      目的:研究疏肝健脾饮对乳腺癌癌前病变裸鼠淋巴管生成途径中COX-2/VEGF通路中VEGF-C、VEGFR-3、COX-2蛋白及MRNA表达的影响,寻找中药复方对于抑制淋巴管生成的作用靶点,探讨疏肝健脾饮及其拆方对乳腺癌癌前病变淋巴管生成的干预作用机制。方法:将120只BALB/c-nude mice裸鼠乳腺原位移植MCF-10AT细胞株,建立癌前病变模型,随机分成6组,模型对照组,疏肝健脾饮组,疏肝理气组,健脾化痰组,活血化瘀组,他莫昔芬组。用药后通过Western-blot、RT-PCR技术检测肿瘤组织中VEGF-C、VEGFR-3、COX-2的蛋白及MRNA表达情况。结果:各用药组VEGF-C、VEGFR-3、COX-2的蛋白及MRNA表达均不同程度减弱,且疏肝健脾饮组减弱明显,与模型对照组相比具有统计学差异(P<0.05)。结论:疏肝健脾饮通过干预VEGF-C、VEGFR-3、COX-2的表达,影响淋巴管的生成,抑制乳腺癌癌前病变。

      Objective: To research the effect on protein and MRNA expression of VEGF-C、VEGFR-3、COX-2 of Lymphangiogenesis in nude mice with precancerous lesions of breast cancer,search the targets about Inhibition of lymphangiogenesis by Chinese herbal compound.Investigate the intervention mechanism of Shuganjianpiyin recipe and its separated recipes on Lymphangiogenesis in nude mice with precancerous lesions of breast cancer?.Methods: 120 BALB/c-nude mice transplanted MCF-10AT cell line on their breast to establish the precancerous lesion model were randomly divided into 6 groups, the model group,the Shuganjianpinyin group,the Shuganliqi group,the Jianpihuatan group,the Huoxuehuayu group,the Tam group.Through Western-blot and RT-PCR Technology test protein and MRNA expression of VEGF-C、VEGFR-3、COX-2 of tumor tissue.Results :Each drug group diminishes in varying degrees in protein and MRNA expression of VEGF-C、VEGFR-3、COX-2,and the group of Shuganjianpiyin recipe slow-up.There was a statistical difference compared with the model control group(P<0.05).Conclusion: Shuganjianpiyin can Influence the formation of lymphatic vessels and control the breast precancerous lesion through intervening the expression of VEGF-C、VEGFR-3、COX-2 .
