      目的 本文通过构建心肌缺血/再灌注损伤(MIRI)大鼠模型,基于细胞凋亡相关通路,观察白金方(乌腺金丝桃、薤白、瓜蒌)对细胞凋亡的影响,进而探讨其对大鼠心肌缺血再灌注损伤的心肌保护作用及相关机制。方法 将3月龄的SD大鼠随机分为空白组、假手术组、模型组、预处理组、白金方不同比例配伍组(乌腺金丝桃:薤白:瓜蒌:1:1:3组、2:1:3组、3:1:3组)。预处理结束后,经开胸手术,结扎冠状动脉左前降支造成心肌缺血,构建大鼠MIRI模型。白金方不同比例配伍组使用不同比例配伍的白金方,对其药物灌胃14天。采用ELISA法测定心肌组织中MDA、SOD含量,比较不同配伍比例的白金方的心肌保护作用。使用荧光定量PCR检测心肌细胞凋亡相关基因(BCL-xl mRNA、BCL-2 mRNA),了解白金方对大MIRI大鼠的心肌保护作用。结果 白金方(乌腺金丝桃:薤白:瓜蒌)1:1:3组、2:1:3组、3:1:3组均能使心肌组织中的氧化应激指标MDA、SOD含量降低。2:1:3组、3:1:3组不同程度增加抑制凋亡基因BCL-xl mRNA、BCL-2 mRNA的表达,来抑制细胞凋亡,其中3:1:3组抗凋亡能力最佳。结论 白金方各配伍组均可不同程度的调节大鼠MIRI过程的凋亡相关基因,上调抗凋亡基因BCL-xl mRNA、BCL-2 mRNA的表达,各组间比较结果提示3:1:3组在调控凋亡相关基因方面具有显著优势,从而发挥其心肌保护作用。

      Abstract: Purpose In this study, we constructed a MIRI rat model and observed the effect of platinum formula (Hypericum allium, Allium macrostemon, Gualsanthis) on cell apoptosis based on apoptosis-related pathways, and then explored its myocardial protective effect on myocardial ischemia-reperfusion injury in rats and its related mechanisms.Methods Three months old SD rats were given white gold prescription in different proportions by intragastric administration for 14 days.After the pretreatment, the left anterior descending branch of the coronary artery was ligation through thoracotomy to cause myocardial ischemia, and then the ligation thread was unwound to construct the rat MIRI model.Subsequently, the contents of MDA and SOD in myocardial tissue were determined by ELISA, and the myocardial protective effects of different compatibility ratios of platinum formula were compared.Fluorescence quantitative PCR was used to detect cardiomyocyte apoptosis-related genes (Bcl-xL mRNA and Bcl-2 mRNA) in order to understand the myocardial protective effect of platinum formula on large MIRI rats. Results Group 1:1:3, group 2:1:3 and group 3:1:3 of white Gold formula (Hypericum allium:Allium macrostemon:Gualsanthis) could reduce the contents of MDA and SOD in myocardial tissue.The expressions of Bcl-xL mRNA and Bcl-2 mRNA of anti-apoptotic genes were increased in the 2:1:3 group and the 3:1:3 group to different degrees to inhibit apoptosis, and the 3:1:3 group showed the best anti-apoptotic ability. Conclusion The apoptosis-related genes in the MIRI process of rats were regulated to varying degrees in the matched groups of platinum formula, and the expressions of anti-apoptotic genes Bcl-xL mRNA and Bcl-2 mRNA were up-regulated, so as to play their myocardial protective effects.
