      目的:研究育阴灵加减方对早发性卵巢功能不全(POI)模型大鼠的影响及其可能作用机制。方法:35%D-半乳糖造模成功的POI模型雌性大鼠40只随机分为模型组、中药高(0.562g/kg)、中(0.281g/kg)、低(0.141g/kg)剂量组和西药组,西药组予戊酸雌二醇溶液0.120mg/kg,第4天加灌醋酸甲轻孕酮片溶液0.960mg/kg,随后两药停用1天,为1个疗程,灌胃4个疗程。未造模的8只雌性大鼠作为空白组。观察大鼠体重、卵巢及子宫系数、卵巢组织的病理变化、血清抗缪勒管激素(AMH)、抑制素B(INHB)、血管内皮生长因子(VEGF)水平,蛋白免疫印迹法检法及荧光定量PCR法测定卵巢组织中Hippo信号通路相关效应因子表达情况。结果:与模型组相比中药高、中剂量组可以升高大鼠体重、卵巢系数、子宫系数(P<0.05);改善卵巢组织结构;升高大鼠血清AMH、INHB、VEGF水平(P<0.05);Hippo信号通路相关效应因子Mst1、Lats2、YAP1、p-YAP、Smad7的表达量明显降低(P<0.05)。结论:育阴灵加减方可能是通过调控Hippo信号通路的表达起到改善POI模型大鼠卵巢功能的作用。

      Objective: To study the effect of Yuyinling Modified Formula on POI model rats and its possible mechanism. Methods: The POI rat model was established by 35% D-galactose, 40 female rats were randomly divided into model group,Chinese medicine high-dose(0.562g/kg),medium-dose(0.281g/kg),low-dose(0.141g/kg) groups and western medicine group. The western medicine group was given estradiol valerate solution 0.120mg/kg, and megalprogesterone acetate tablet solution 0.960mg/kg, on the 4th day, the two drugs were stopped for 1 day, which was a course of treatment, gavage was administered for 4 courses. Eight unmodeled female rats were used as a blank group. Body weight, ovarian and uterine coefficient, pathological changes of ovarian tissue and serum AMH, INHB and VEGF levels were observed. Western Blot and RT-PCR were used to determine the expression of Hippo signaling pathway in ovarian tissue. Results: Compared with model group, body weight, ovarian coefficient and uterus coefficient of rats in high and medium dose group of Chinese medicine could be increased (P<0.05); Ovarian tissue structure is improved; Serum AMH, INHB and VEGF levels were significantly increased (P<0.05). The expression levels of Hippo signaling pathway related effect factors Mst1, Lats2, YAP1, p-YAP and Smad7 decreased significantly. Conclusion: Yuyinling Modified Formula may improve the ovarian function of POI model rats by regulating the expression of Hippo signaling pathway.
